I have no idea what the Greensboro contest is. And I have no idea what the difference would be between actual words or manuscript-format words, unless the latter is some bastardized form of the New Math -- in which case I'll side with actual words. Especially for a contest.

If you keep your actual word count (i.e., the word count your word processor gives you) within the 4000-word limit, you won't have any problems.
